Aspartate aminotransferase (AspAT) of Pseudomonas aeruginosa has been purified in a homogenous form, about 100-fold, using conventional chromatographic methods. In order to characterize its biochemical properties, several chemical agents were tested, such as thiol reagents, heavy metals, detergents and organic solvents, for their ability to inhibit the enzyme activity. Only sulfhydryl compounds and heat denaturation were able to completely inhibit the enzyme activity. In addition, a kinetic study was carried out using N-methyl-aspartate, aspartate, 2-oxoglutarate, and oxaloacetate as substrate. The kinetic parameters were calculated by fitting the data to Michaelis-Menten kinetics. The Km values for the substrates were 5.53 +/- 0.23 mM for N-methylaspartate, 0.735 +/- 0.038 mM for aspartate, 0.60 +/- 0.03 mM for 2-oxoglutarate and 0.524 +/- 0.034 mM for oxaloacetate. The enzyme also has a broad pH range for activity, being active between pH 4.5 and 10.0.
