To investigate whether BIRC5 is involved in the apoptosis pathway of oral squamous cell carcinoma (OSCC) cells and its association with the sensitivity of OSCC cells to cisplatin. BIRC5 expression in 25 OSCC and paired normal oral epithelium tissues was determined by reverse transcription polymerase chain reaction and western blotting. OSCC cell line Tca8113 cells and normal human gingival fibroblast cell line HGF-1 cells were infected with BIRC5 small interfering RNA (siRNA) or BIRC5 overexpressing vector. Cell viability was measured by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay. Cells were also treated with cisplatin. Cell apoptosis was evaluated using an Annexin V-FITC/propidium iodide (PI) assay. Immunofluorescence staining was performed to detect caspase-3 activity. BIRC5 expression was significantly lower in OSCC tissues compared to that in the normal tissue. Moreover, down-regulation of BIRC5 in Tca8113 cells resulted in enhanced sensitivity to cisplatin-induced apoptosis. This effect was also observed in HGF-1 cells after BIRC5 overexpression. Furthermore, up-regulation of BIRC5 in Tca8113 cells significantly decreased the sensitivity of Tca8113 cells to cisplatin. The expression of Fas, FADD and caspase-3 were decreased in Tca8113 cells after BIRC5 down-regulation. BIRC5 deletion in OSCC cells enhances the sensitivity of OSCC cells to cisplatin. BIRC5-induced cisplatin resistance in OSCC cells is associated with the enhancement of Fas/Fas ligand (FasL) pathway.Q:
